深圳市19例登革热患者血清代谢组分析

Analysis of the serum metabolome of 19 dengue fever patients in Shenzhen

摘要: 目的 对登革病毒(dengue virus,DENV)感染者和健康人群进行代谢组学对比分析,探究登革热患者血清的代谢组学变化,通过生物信息学方法进行差异分析和通路分析,了解宿主对病毒的代谢反应,寻找可能的生物标志物。方法 以2023年6月至10月就诊于深圳市第三人民医院的19例登革热患者作为研究对象,同时以11例健康人群作为对照,采集其血清,进行非靶向代谢组学分析。通过生物信息学方法进行代谢物表达差异分析和通路分析,筛选差异表达的代谢物,寻找病毒感染影响的宿主代谢通路。结果 总体主成分分析(principal component analysis, PCA)和相对标准差(relative standard deviation,RSD)显示生物重复样本的定量结果符合统计学一致性。相比健康人群,登革热患者的血清代谢物共有114种代谢物展现出明显的表达差异(P<0.05,差异倍数≥1.5或≤2/3)。对差异代谢物进行人类代谢数据库(human metabolome database,HMDB)注释分类结果显示差异代谢物主要集中在脂类代谢物上。结论 登革热患者的血清代谢组学特征与健康个体显著不同,其主要特征是脂质代谢的改变。这些结果表明登革病毒感染可能会改变宿主的脂代谢途径,为登革病毒感染的致病机制研究提供理论基础,有助于登革热的诊断和预后评估。

Abstract: Objective To explore the metabolomic changes in the serum of dengue patients through comparative analysis of the metabolome of patients infected with dengue virus (DENV) and healthy people. Differential analysis and pathway analysis were performed to understand the host's metabolic response to the virus and to search for possible biomarkers. Method 19 dengue fever patients who visited Shenzhen Third Peoples Hospital from June 2023 to October were used as the research subjects, and 11 healthy people were used as controls. Their serum was collected for non-targeted metabolomic analysis. Use bioinformatics methods to conduct differential analysis and pathway analysis to screen differentially expressed metabolites and find host metabolic pathways affected by viral infection. Result Overall principal component analysis (PCA) and relative standard deviation (RSD) showed that the quantitative results of biological replicate samples were statistically consistent. Compared with the normal population, the serum metabolites of dengue patients in the experimental group were significantly different. A total of 114 metabolites showed significant expression differences(P<0.05,Fold Change≥1.5 or ≤2/3). The HMDB annotation classification of differential metabolites showed that the differential metabolites were mainly concentrated on lipid metabolites. Conclusion The serum metabolomic profile of dengue patients significantly differs from that of healthy individuals, primarily characterized by alterations in lipid metabolism. These findings suggest that dengue virus infection modifies the host's lipid metabolism pathway, providing a theoretical foundation for understanding the pathogenesis of dengue virus infection and contributing to the diagnosis and prognosis of dengue fever.
