等温扩增技术检测人乳头瘤病毒的研究进展

Progress in the detection of human papillomavirus by isothermal amplification technology

摘要: 宫颈癌是全球第四大女性癌症, 几乎所有宫颈癌病例都与人乳头瘤病毒(human papillomavirus,HPV)感染有关,因此,HPV感染的早期发现对预防宫颈癌至关重要。WHO建议将HPV DNA检测作为主要筛查方法,等温扩增技术相较于变温扩增技术操作更简便,反应更高效,对实验条件,操作人员的要求更低,且经济成本更低,因此,更适用于HPV的检测。本文比对分析了用于HPV检测的等温扩增技术方法,常用的扩增产物判读方法及等温扩增技术与其他技术(如微流控芯片技术、CRISPR-Cas技术、智能手机检测和横向流动试纸条)结合检测的应用现状。对这些方法和技术进行优缺点分析,对其所适合的应用场景进行阐述,总结等温扩增技术存在的不足并对其发展前景进行展望。

Abstract: Cervical cancer is the fourth most common female cancer in the world. Almost all cases of cervical cancer are associated with human papillomavirus (HPV) infection, so early detection of HPV infection is essential for cervical cancer prevention. The World Health Organization recommended HPV DNA testing as the primary screening method. Isothermal amplification technology is more suitable for HPV testing because it is easier to operate and more efficient than variable temperature amplification technology, which requires less laboratory conditions, less operator effort, and lower economic costs. This paper comparatively analyses the advantages and disadvantages of isothermal amplification techniques used for HPV detection, commonly used amplification product readout methods and the current status of the application of isothermal amplification techniques in combination with other techniques (e.g., microfluidic chip technology, CRISPR-Cas technology, smartphone assay, and lateral flow dipstick) for detection of HPV. At last, the advantages and disadvantages of these methods and techniques are analyzed, their suitable application scenarios are elaborated, and the shortcomings of isothermal amplification techniques are summarized and the prospects for their development are outlooked.
